O-039 Novel sperm centrosome function, structure, and composition during fertilization

چکیده

Abstract Sperm centrioles play a critical role in sperm differentiation and embryo development, they are implicated infertility miscarriages. However, the precise way function inherited during fertilization mammals remains unresolved. A key reason for this lack of understanding is that many proteins only transiently present centriole because process known as Centrosome Reduction exhibit dramatic species-specific adaptions. It was long widely accepted structural degeneration functional inactivation occur at one or two mammals, resulting vestigial remnant. This led to belief human spermatozoa have (the Proximal Centriole, PC). While happens mice, our studies suggest present, both essential non-murine mammals. Distal Centriole (DC) has highly atypical structure composition, while PC modestly modified. These findings us new model: Remodeling, which states preexisting spermatid centrioles, along with their pericentriolar material (PCM), go through compositional transformation remodeled PCM functions dynamic basal complex (DBC) spermatozoon precursor embryonic centrioles. crucial mystery remaining is: what centrioles' functions? To answer question, we study remodeling infertile men rabbit mutants partially disabled Trial registration number -

Receptor function of mouse sperm surface galactosyltransferase during fertilization

Past studies from this laboratory have suggested that mouse sperm binding to the egg zona pellucida is mediated by a sperm galactosyltransferase (GalTase), which recognizes and binds to terminal N-acetylglucosamine (GlcNAc) residues in the zona pellucida (Shur, B. D., and N. G. Hall, 1982, J. Cell Biol. 95:567-573; 95:574-579). We now present evidence that directly supports this mechanism for g...

Speriolin is a novel human and mouse sperm centrosome protein.

BACKGROUND Oocytes in humans, mice and other mammals lack identifiable centrioles. The proximal centriole brought in by the fertilizing sperm in humans and most other mammals appears to gives rise to the centrioles at the spindle poles in the zygote, and is believed to indicate that centrioles are inherited through the paternal lineage. However, both the proximal and distal sperm centrioles deg...

O-5: Identification of Novel ImmunodominantEpididymal Sperm Proteins Using CombinatorialApproach

Background: Alteration in the protein signatures of functionally immature testicular spermatozoa occurs during their journey through the epididymis. This leads to acquisition of sperm domain specific functions essential for successful fertilization. Epididymal sperm proteins are preferred targets for immunocontraception as well as in elucidating the causes of infertility. The Background of the ...

Function of sperm chromatin structural elements in fertilization and development.

Understanding how DNA is packaged in the mammalian sperm cell has important implications for human infertility as well as for the cell biology. Recent advances in the study of mammalian sperm chromatin structure and function have altered our perception of this highly condensed, inert chromatin. Sperm DNA is packaged very tightly to protect the DNA during the transit that occurs before fertiliza...

CRISP1 as a novel CatSper regulator that modulates sperm motility and orientation during fertilization

Ca(2+)-dependent mechanisms are critical for successful completion of fertilization. Here, we demonstrate that CRISP1, a sperm protein involved in mammalian fertilization, is also present in the female gamete and capable of modulating key sperm Ca(2+) channels. Specifically, we show that CRISP1 is expressed by the cumulus cells that surround the egg and that fertilization of cumulus-oocyte comp...
